Exploration of chemical space for drug discovery by database generation
Small molecule drug discovery requires constant innovation The exhaustive enumeration of the 26.4 million possible small organic molecules up to 11 atoms of C, N, O, F in the database GDB shows that 99.8 percent of the small molecule chemical space has yet to be explored. Guided by virtual screening tools, a proof-of-principle study shows that novel bioactive ligands from GBD can be readily ide...
متن کاملTethering small molecules to a phage display library: discovery of a selective bivalent inhibitor of protein kinase A.
There is much current interest in developing fragment-based strategies for targeting protein surfaces and active sites.1 Toward this goal, we describe a new fragment-based bivalent ligand selection methodology that allows for the discovery of protein surface-targeted cyclic peptides that are steered by an active site binding small-molecule ligand (Figure 1). This approach allows for coupling hi...
متن کاملA chemogenomic approach to drug discovery: focus on cardiovascular diseases.
References to individual protein targets and bioactive small molecules associated with cardiovascular diseases can be found in multiple bibliographic sources. From mining these sources, a highly curated list of 214 cardiovascular targets was collected and organised using functional classification schemes for the main protein families of therapeutic relevance, namely, enzymes, G-protein-coupled ...
متن کاملIdentification of a Novel Tumor-Binding Peptide for Lung Cancer Through in-vitro Panning
Tumor-targeted therapies are playing growing roles in cancer research. The exploitation of these powerful therapeutic modalities largely depends on the discovery of tumor-targeting ligands. Phage display has proven a promising high throughput screening tool for the identification of novel specific peptides with high binding affinity to cancer cells. In the present study, we describe the use of ...
